Manning limited his workout to protect his throwing arm, but head coach Tony Dungy said he did not believe Manning would miss Sundays game against New England.
"He felt a twinge in there during our walkthrough this morning, so we took a lot of the work off of him," Dungy said.
Dungy said Manning was to have the MRI later Wednesday, but it was not clear when there would be word of the results.
With Manning limited, Brock Huard -- the only other quarterback on the Colts roster -- worked with the first team.
Manning, having the best season of his six-year NFL career and a leading contender for the MVP award, has never missed a start.
Hes on pace for a career-high in yards passing, a career-low in interceptions and his 3,105 yards and 67.4 percent completion percentage lead the NFL.
Mannings troubles are the latest chapter in an injury-plagued season for the Colts.
